小鼠口服两性霉素B脂质卷和胶束溶液后的组织分布

Drug tissue distribution after oral delivery of amphotericin B cochleate and amphotericin B micellar solution in mice

摘要 两性霉素B (AmB)属多烯类广谱抗真菌抗生素,是第一个用于深部真菌感染的药物.但AmB在水中溶解度低,口服无效,目前临床应用较多的为冻干粉针剂,含去氧胆酸钠作为增溶剂,加水溶解可形成胶束,注射给药.但因严重的不良反应尤其是肾毒性,极大地限制了它的临床应用.上市的AmB脂质给药剂型(脂质体、胶态分散体、脂质复合物)[1]能显著减少AmB的毒性,但均需注射给药,且价格昂贵.
