摘要
目的:探讨新生鼠缺氧缺血性脑损伤中 c-fos 和 c-jun 表达的意义。方法:Wistar 大鼠18只分为两组,实验组12只,对照组6只,制备脑缺氧缺血动物模型,用 RT-PCR 和斑点杂交方法检测新生鼠缺氧缺血性脑损伤后 c-fos 和 c-jun 在神经细胞中的表达。结果:c-fos 和 c-jun的表达在不同时间段具有显著性差异(P<0.001),其中72小时组和96小时组的表达比24小时高,缺氧缺血后24~96小时,c-fos 和 c-jun 在躯体感觉皮质区,海马回 CA3区和基底节区持续表达,与对照组相比有统计学意义(P<0.01);c-fos 和 c-jun 的表达在不同脑区也具有显著性差异(P<0.01),海马区与基底节区大于皮质区。结论:c-fos 和 c-jun 的表达增加不仅是脑缺氧缺血的结果,而且可能参与了对神经细胞损害的过程。
Objective:To investigate the relationship between the expression of c-fos as well as c-jun and cerebral hypoxia-ischemia in newborn rats.Methods:18 Wistar rats made the model of hy-poxia-ischemia in experiment were divided into two groups,the group of experiment used 12 rats.Thecontrol group were 6 rats.The Rt-PCR and blots hybridization were used in experiment.Results:Therewere significance variation between the expressionoof c-fos and c-jun in different time,(P<0.01,which the groups of 72 hours and 92 hours were higher than the groups of 24 hours)in different aeres(P<0.01,which the areas of CA3 of dental grays and basal ganglia were bigger than the areas of soma-tosenssr).Conclusion:these results indicated that the increase in expression genes c-fos,c-jun couldbe induced by cerebral hypoxia-ischemia,and the genes may be one of the gene induced neuron cell ap-optosis.
