摘要
目的 观察中华眼镜蛇膜毒素 (CT)和131I与抗鼻咽癌 (NPC)单克隆抗体BAC5的偶联物 ,对NPCCNE 2细胞微球的抑制或破坏作用。方法 采用氯胺 T法制备131I BAC5,并用异型双功能交联剂 (SPDP)偶联CT BAC5,分别或联合应用于NPCCNE 2细胞微球 ,观察微球生长的体积变化率和评价其受抑制或受破坏的情况。结果 CT和BAC5的偶联率为 32 .4% ,与对照组比较 ,CT对微球的生长无明显抑制作用 (P >0 .0 5 ) ,CT BAC5有明显的的抑制作用 (P <0 .0 5 ) ,131I BAC5、131I BAC5+CT BAC5有非常明显的破坏作用 (P <0 .0 1)。结论 肿瘤多细胞微球是一种用于体外研究肿瘤治疗效果的理想模型之一。CT BAC5和131I BAC5联合免疫导向治疗NPC比单项治疗效果更好。
Objective To develop a new specific therapeutic method for treating nasopharyngeal carcinoma(NPC) and to evaluate its effects on an in vitro tumor spheroid model. Methods The human NPC cell line CNE 2 spheroids were grown in culture medium, on a thin layer of 0.75% agarose.The average volume of the spheroids was estimated by 1/2 (short dimension) 2 ⅹ(long dimension). Anti NPC BAC 5 MAb was labeled with 131 I using chloramine T method.Cytotoxin from Chinese cobra (CT) was cojugated with BAC 5 by heterobifunctional cross linking reagent(SPDP).The spheroids were treated with either 131 I BAC 5 or CT BAC 5 separately or with both of them together by incubating in culture medium. Results The conjugated rate of BAC 5 and CT was 32.4%.CT BAC 5 significantly inhibited the growth of CNE 2 multicellular spheroids comparing with control agent(normal saline) ( P <0.05) and 131 I BAC 5 did more than CT BAC 5 ( P <0.01) .The combined 131 I BAC 5 with CT BAC 5 treatment showed even stronger destructire effect on the spheroids (vs.normal saline, P <0.01). ConclusionsTumor multicellullar spheriods is one of ideal models for evaluating therapeutic effects on tumor in vitro.The effect of immunotherapy with combined 131 I BAC 5 with CT BAC 5 is better than either of them working separately.
