摘要
目的探讨PTEN、AKT2、NF-кB在肺癌组织表达及临床意义。方法随机取62例肺癌组织作为肺癌组与20例肺癌远端(>5cm)肺组织为癌旁组,采用免疫组化法与RT-PCR方法检测组织中PTEN、AKT2、NF-кB蛋白的表达,并分析其相关性和临床意义。结果肺癌组织中AKT2、NF-кB的阳性表达率分别为57.89%和79.00%,显著高于癌旁组(15.00%和50.00%);肺癌组PTEN阳性表达率为50.00%,显著低于正常组(90.00%),差异有统计学意义(均P<0.05)。RT-PCR显示肺癌组织中AKT2、NF-кB的mRNA表达量显著高于癌旁组,而PTEN的mRNA表达量显著低于正常组,组间比较差异有统计学意义(P<0.05)。PTEN与NF-кB的表达在肺癌组织不同分化程度方面差异显著;PTEN、AKT2、NF-кB的表达在不同的临床分期与有无淋巴结转移方面,差异有统计学意义(均P<0.05);PTEN、AKT2、NF-кB的表达与患者的年龄、性别、组织学类型等无关(均P>0.05)。肺癌组织中PTEN的表达与AKT2、NF-кB呈负相关(均P<0.05),而AKT2与NF-кB呈正相关(均P<0.05)。结论在肺癌组织中,PTEN呈低表达,AKT2、NF-кB呈高表达,PTEN、AKT2、NF-кB的表达与肺癌发生发展关系密切,可以作为临床诊治肺癌的重要参考指标。
Objective To study expression and clinical significance of PTEN,AKT2,NF-кB in tissues of lung canc-er.Methods The expression of PTEN,AKT2,NF-кB in 62 cases of lung cancer and 10 cases of normal lung tissues were detected by immunohistochemistry.Results The positive expression rate of AKT2,NF-кB in the lung cancer tis-sues are57.89%,79.00%,significantly higher than in normal lung tissue (15.00%,50.00%);Positive expression rate of PTEN in lung cancer group is 50.00%,significantly lower than normal lung tissues (90.00%),(all P 〈 0.05). AKT2NF-k B mRNA expression quantity of lung cancer group respectively are (59.43&#177;1.29),(79.19&#177;1.29),sig-nificantly higher than in normal lung tissue group (9.91&#177;1.85),(44.28&#177;2.49);Lung cancer group of PTEN mRNA expression quantity (22.31&#177;2.77)was significantly lower than normal lung tissue (59.64&#177; 1.36);(all P 〈0.05). The expression of PTEN and NF-кB seem different in lung cancer tissue differentiation degree has significant differ-ence;the expression of PTEN,AKT2,NF-кB seem in different clinical stage and the presence of lymph node metasta-sis,the difference was statistically significant (all P 〈0.05).Low differentiation,lymph node metastasis in lung cancer tissues and III-IV,PTEN show lower expression and expression of AKT2,NF-кB seem high;PTEN,AKT2,the ex-pression of NF-B seem with the patient&#39;s age,gender,histologic type,etc.(all P〉0.05).Conclusion In lung cancer tis-sues,PTEN is has low expression,AKT2,NF-кB seem has high expression,the expression of PTEN and AKT2,NF-кB seem closely associated with lung cancer occurrence and condition of development,can guide clinical diagnosis and treatment.
