操作胁迫对云纹石斑鱼肝脏抗氧化和鳃Na+-K+ATPase活力的影响 被引量:20

Effects of handling stress on activities of antioxidant enzymes in liver and Na^+-K^+ ATPase in gill of Epinephelus moara

摘要 研究了操作胁迫(惊扰)对云纹石斑鱼(Epinephelus moara)肝脏抗氧化能力和鳃中钠-钾ATP酶(Na+-K+ATPase)活力的影响。在胁迫处理后,鱼体肝脏中抗氧化酶活力受到不同程度的影响。超氧化物歧化酶(SOD)和过氧化氢酶(CAT)具有相似的变化规律,其在处理后快速达到活力最高值,然后逐渐下降到与处理前无显著性差异(P>0.05);谷胱甘肽转移酶(GST)活力也在处理后升高,但一直与处理前无显著性差异(P>0.05);肝脏中VC含量在处理后显著增高,并最终恢复到处理前水平;而Na+-K+ATPase活力一直显著低于处理前水平(P>0.05)。研究表明,操作胁迫会通过氧化应激对云纹石斑鱼体造成损伤,而在胁迫应激反应中,SOD和CAT可以作为有效的生物指示剂用于胁迫状态的检测。 In aquaculture, fish are regularly exposed to a variety of acute and chronic stressors. Such stressors can induce physiological responses with adverse effects on growth, reproduction, immune response and fillet quality. Understanding and monitoring biological mechanisms underlying stress responses will facilitate alleviating their negative effects through selective breeding and changes in management practices, resulting in improved animal welfare and production efficiency. In the present study, effects of handling stress ( physical disturbance) on activities of antioxidant enzymes in liver and Na + -K ~ ATPase in gill of Epinephelus moara were studied. After being subjected to handling stresses, activities of antioxidant enzymes were influenced in different degrees. SOD and CAT showed similar changes, their activities increased significantly after exposure to stressors and then dropped to levels before treatment ( P 〈 0.05 ). GST activities insignificantly increased after stress ( P 〉 0.05 ). Concentrations of VC in liver also increased significantly after stress, and then dropped. But activities of Na + -K + ATPase maintained a significantly low level throughout the whole process of stress ( P 〈 0.05 ). The present study suggested that handling stress would damage E. rnoara by oxidant stress, and SOD and CAT could be used as biomarkers in monitoring of stress status.
