茶叶水提取物诱导LoVo细胞凋亡的差异 被引量:5

摘要 目的探讨几种绿茶、红茶和乌龙茶水提取物诱导大肠癌LoVo细胞凋亡的作用及其差异性。方法用台盼蓝拒染法显示二种绿茶、二种红茶和二种乌龙茶水提取物对LoVo细胞的细胞毒作用;用HE染色、琼脂糖凝胶电泳观察上述处理因素对LoVo细胞在形态学和生化方面的影响,通过流式细胞仪对LoVo细胞的凋亡进行定量分析。结果台盼蓝拒染法显示不同品种的茶叶水提取物对LoVo细胞的生长具有抑制作用,并具有时间和剂量效应关系,这种细胞毒效应在不同茶叶水提取物之间存在差异。HE染色显示几种茶叶水提取物皆能诱导LoVo细胞的凋亡,出现典型的凋亡细胞形态学改变:细胞核固缩、碎裂等。琼脂糖凝胶电泳上呈现典型的“梯状”带,流式细胞仪定量分析表明不同茶叶水提取物所致的凋亡率不同。结论不同品种的茶叶水提取物可诱导大肠癌LoVo细胞的凋亡,其作用强弱依次为绿茶、红茶和乌龙茶,茶叶诱导LoVo细胞凋亡除主要由其中的儿茶素介导外,可能还有其他成分的参与。 AIM To investigate the difference in the induction of apoptosis of LoVo cells, a human colon carcinoma cell line, by different tea aqueous extracts. METHODS After exposing LoVo cells to different tea aqueous extracts including two species of green tea, black tea, and oolong tea, respectively, the growth suppression was determined by trypan blue exclusion and the apoptotic alterations in morphology and biochemistry were detected by hematoxylin-eosin (HE) staining and gel electrophoresia. Additionally, the apoptotic rates were quantified by flow cytometry with PI staining. RESULTS Treatment of LoVo cells with different tea aqueous extracts suppressed the growth of LoVo cells in dose. and time-dependent manner. There was marked difference in degree of the cytotoxicity. Morphologic analysis showed apoptotic alterations such as chromatin condensation and nuclear fragmentation. Agarose gel electrophoresis of the soluble DNA exhibited the presence of oligonucleosomal-sized fragments, a characteristic, considered as a hallmark of apoptosis. In addition, quantitative assessment of apoptosis-associated hypodiploid by FCM showed different apoptotic rates of LoVo cells caused by different tea aqueous extracts. CONCLUSION Different tea aqueous extracts have various effects on inducing apoptosis of LoVo cells. The order of the effects from strong to weak is the green tea, black tea, and oolong tea aqueous extracts, respectively. There are possibly other components in tea taking part in the induction of apoptosis of LoVo cells besides catechins.
