摘要
目的:筛选纯化瓦松总黄酮的树脂型号及最佳工艺。方法:分别通过动态、静态吸附的方法,以解吸率为主要指标考察各因素对瓦松总黄酮大孔吸附树脂纯化工艺的影响。结果:DM301型大孔树脂纯化瓦松总黄酮效果最好,最佳工艺条件为:上样液浓度2.65 mg·ml-1,吸附速率3 BV·h-1,50%乙醇为洗脱液,8倍量树脂柱体积洗脱,上样液pH2时效果最好,总黄酮含量达到54.11%。结论:DM301型大孔吸附树脂纯化瓦松总黄酮操作简单、安全、成本低廉、有较高的应用价值。
ORJECTIVE To select the best resins model and the best craft for purification of total flavonoids from Orostachys fimbriatus. METHODS The technological conditions of purification were examined by the static adsorption and desorption ex- periments, and taking the desorption rate as the main index. RESULTS DM301 type macroporous resin was the best agent for purification of total flavonoids from Orostachys fimbriatus. The optimal conditions were as follows, the column liquid concen- tration 2. 65 mg·m1-1 , adsorption rates 3BV, eluting with 50% ethanol and 8 multiples of column volumn was the best, pH value of sample was 2. In this condition, the purity of total flavonoids was up to 54. 11%. CONCLUSION DM301 macro- porous adsorption resin is safe , economical and easy to use and can therefore be promisingly applied to purify total flavonoids from the Orostach ys f imbriatus.
