摘要
目的:探讨黄芪皂苷、黄芪多糖不同浓度水平等比配伍对正常培养和脂多糖(LPS)诱导培养的小鼠腹腔巨噬细胞免疫活性的影响。方法:制备小鼠腹腔巨噬细胞悬液,依次进行正常培养和LPS诱导培养,采用MTT微量比色法检测吸收值,并于培养结束前取各孔培养液上清,检测一氧化氮(NO)含量。结果:对正常培养的腹腔巨噬细胞,与空白对照组比较,配伍浓度500,200μg·ml-1能明显促进细胞增殖,浓度为500μg·ml-1时,Q值大于1.15,浓度小于200μg·ml-1时,Q值均小于0.85,且浓度在500,200,40μg·ml-1能明显促进细胞的NO释放;对LPS诱导培养的腹腔巨噬细胞,与模型对照组比较,配伍浓度大于200μg·ml-1对细胞增殖具有显著促进作用,低于40μg·ml-1有明显抑制作用,且浓度为40~500μg·ml-1时,Q值大于1.15,浓度为1.6~8μg·ml-1时,Q值小于0.85,同时各配伍浓度对NO的释放量均表现出明显抑制作用。结论:黄芪组分配伍对正常培养和LPS诱导培养的小鼠腹腔巨噬细胞体外增殖均表现为高浓度促进,低浓度抑制,高浓度配伍为协同效应,低浓度配伍为拮抗效应,且其对NO分泌的影响与细胞所处状态及药物浓度相关。
ORJECTIVE To explore the influence of compatibility of astrageloside (AS) and Astragalus polysaccharide (APS) under different concentrations on immune activity in the normal and LPS-induced cultured macrophages of mice. METIt- Oils Preparation of peritoneal macrophage suspension of mice, the normal culture and LPS induced medium were carried on in turn, the absorption value of maerophage was detected by MTT microcolorimetry after 48h and nitric oxide (NO) detected in each hole culture supernatant. RESULTS Compared the normal peritoneal macrophages of mice in vitro with the blank control group, Astragalus group compatibility had obvious promoting effect on cell proliferation at high concentrations 500, 200μg· m1-1. And when the concentration at 500 μg· ml-1, Q value was greater than 1.15, when the concentration was less than 200μg·ml-1, Q values were less than 0. 85. Component compatibility at the concentration of 500,200, 40μg·ml-1 had signifi- cant effect on cells to promote the release of NO. Compared the LPS induced peritoneal macrophages of mice in vitro with the model group, when the compatibility concentration greater than 200 μg·ml-1 showed significant effect on cell proliferation, when the concentration less than 40μg·ml-1 1could significantly inhibit the cell growth. And when the concentration was greater than 40μg·m-l 1, the Q value was greater than 1.15; concentration was less than 8μg·ml-1 , Q values less than 0. 85, the compatibility could all inhibit NO content in different concentration. CONCLUSION The effect on proliferation of component compatibility are characterized by that at high concentrations it could promote the cell growth contrary to low concentrations, and high concentrations showed synergies contrary to low concentrations, and that is the NO content dependent on cell state or drug concentration in cells.
