不同剂量黄芪注射液对人肾小管上皮细胞黏附分子CD146表达的影响 被引量:3

The Exprsession of CD146 of Human Proximal Renal Tubular Epithelial Cells Cultured in High Glucose Condition Influenced by Different Concentrations of Astragalus Injection

摘要 目的:探讨不同剂量黄芪注射液对人肾小管上皮细胞(HK2)黏附分子CD146表达的干预作用。方法:将传代HK2等分为5组,进行培养处理:正常对照组(LG组,只加5.5mmol/L D-葡萄糖),高糖组(HG组,加入25mmol/L D-葡萄糖),黄芪注射液干预组:在HG组中加入不同剂量黄芪注射液,加入2μg/ml者为低剂量组(AML组)、加入20μg/ml者为中剂量组(AMM组)、加入200μg/ml为高剂量组(AMH组)。观察37℃培养24h、48h、72h后,各组HK2形态变化、分别收集各组培养细胞,用Western Blotting检测其CD146蛋白表达。结果:HG组较LG组HK2变长,呈梭形,细胞间连接疏松,间隔增大;黄芪注射液干预各组,细胞形态均较HG组变小变圆,且连接逐渐紧密,AMH组呈堆积生长。HG组CD146表达均较LG组明显增加(P<0.01),干预24h、48h、72h,AMM组、AMH组与HG组比较,CD146表达均明显减少(P<0.05或P<0.01),AML组干预培养72h,CD146的表达亦明显减少(P<0.01)。结论:黄芪注射液改善高糖引起的HK2形态改变和降低其CD146表达,对防治DN有积极作用。 Objective:To study the interference on the expression of adhesion molecules CD146of human renal tubular epithelial cells(HK2)by the intervention of different doses of astragalus injection.Method:HK2were divided into five groups:normal control group(LG group,just add 5.5mmol/L D-glucose),high glucose(HG group,add 25mmol/L D-glucose),astragalus injection intervention groups divided by different concentrations of the HG group on the basis of injection,adding 2μg/ml by low-dose group(AML group),adding 20μg/ml dose group(AMM group),adding 200μg/ml for the high-dose group(AMH group).Cultured at 37℃for 24h,48h,72h,HK2morphological changes were in each group,and cultured cells were collected at each different time.CD146protein expression was detected by western bloting.Results:HK2cultured for 72hin intervention condition,cells of HG group became fusiform than LG group,the intercellular connections loosed and space increased;in the intervention groups,the cells became round and smaller than LG group,and stacked growth.Compared with LG group,intervented for 24h,48h and 72h,the expression of CD146of HG group was significantly increased(P〈0.01),the AMH and AMM group were also significantly reduced(P〈0.01),while the expression of CD146of AML group was significantly reduced after intervention for 72h(P〈0.01).Conclusion:High glucose can stimulate an increase in CD146expression of HK2,differentdoses of astragalus injection can reduce CD146expression and connected with the intervention time and dose.
