牦牛TLR4基因的克隆测序及序列分析 被引量:6

摘要 旨在了解牦牛天然免疫受体TLR4基因特点。根据GenBank已公布的牛TLR4基因序列,分3段设计引物,测序、拼接克隆序列,并对所得序列进行生物信息学分析和预测。基因序列分析表明,克隆得到牦牛TLR4基因编码区全长2 807bp,开放阅读框2 526bp,编码氨基酸841个,N端含有27个氨基酸组成的信号肽,分子质量96.009 8ku,理论等电点为6.35。蛋白预测结果表明,TLR4编码蛋白整体表现为亲水性。同源性分析表明,10条序列当中牦牛与野牦牛、普通牛、绵羊等物种具有较高的同源性,在系统发育树中距离最近。说明TLR4基因序列具有较高的保守性。 This study aims to understand the characteristics of the yak innate immune receptors TLR4 gene.According to the published cattle TLR4 gene sequence in the GenBank,the full gene sequence was divided into three fragments and relevant primers were designed for gene cloning,sequencing and bioinformatics analysis.The results of sequence analysis showed that the length of TLR4 cDNA was 2 807bp,coding a 2 526bp open reading frame(ORF).It encodes a peptide of 841amino acids with a N-terminal signal peptide containing 27amino acids.Its calculated molecule mass is 96.009 8ku and PI is 6.35.The results of protein prediction showed that TLR4 protein overall displayed as a hydrophilic protein.Sequence homology analysis among 10sequences showed that yak had higher identity with wild yak,cattle,sheep and other species,they had the nearest distance in the phylogenetic tree. The results show that TLR4 gene sequence is very conserved in mammals.
