摘要
目的:研究模拟失重对前成骨细胞MC3T3-E1内钙离子浓度的影响。方法:利用回转器模拟失重条件培养前成骨细胞MC3T3-E1 48h。然后将细胞用Fluo-3AM标记,使用激光共聚焦显微镜和流式细胞仪检测细胞内钙离子浓度。结果:模拟失重条件培养后,细胞内钙离子浓度检测显示,与对照组相比,模拟失重组细胞内钙离子浓度降低。流式细胞仪结果显示,与对照组相比,模拟失重组细胞的荧光阳性率显著降低,提示模拟失重可以抑制细胞活性。结论:模拟失重导致前成骨细胞MC3T3-E1内游离钙离子浓度降低,并可能抑制细胞活性。
Objective:To investigate the effect of simulated microgravity on intracellular free Ca2+ concentration in MC3T3-E1pre-osteoblasts.Methods:After culturing in the condition of simulated microgravity created by clinostat rotation for 48hours,MC3T3-E1pre-osteoblasts were dyed by Fluo-3AM,and then the fluorescence intensity and positive rate of fluorescence were measured by the laser scanning confocal microscope and flow cytometry.Results:Compared with control group,decreases of the fluorescence intensity and positive rate of fluorescence were induced by simulated microgravity environment.Conclusion:The intracellular free Ca2+concentration in MC3T3-E1pre-osteoblasts and the activity of the cells could be reduced by simulated weightlessness.
