摘要
目的研究低频超声联合不同抗生素对表皮葡萄球菌(Staphylococcus epidermidis)RP62A生物被膜(biofi lm,BF)渗透性的影响。方法体外建立成熟RP62A BF模型,用扫描电镜(scanning electron microscope,SEM)确认并观察BF的形态结构;平板计数法计算活菌总量;建立生物膜抗生素渗透模型,低频超声联合干预治疗1h、3h,测量抑菌环直径大小。结果低频超声显著增强了庆大霉素和头孢噻肟对表皮葡萄球菌生物膜的渗透性,头孢噻肟联合超声组(1h、3h)和庆大霉素联合超声组(3h)与CB对照组相比均有显著差异(P<0.05);当超声延长至3h后,万古霉素的渗透性略有增强。结论低频超声可不同程度增强抗生素对表皮葡萄球菌生物膜的渗透性。
Objective To study the effect of low-frequency ultrasound combined with antibiotics on penetration of S. epidermidis RP62A strain biofilm (BF). Methods Mature biofilm model of S. epidermidis RP62A strain was established, the BF structures were confirmed and observed by scanning electron microscope(SEM); viable counts were determined by drop plating after suspending the biofilm cells; the penetration of antibiotics through the biofilms was established, after lh and 3h with ultrasonication interventions, the diameter of zones of growth inhibition were measured. Results Low-frequency ultrasonic significantly increased the transport of gentamicin and cefotaxime through the biofilm, cefotaxime combined with low-frequency ultrasound(1h and 3h) and gentamicin combined with low-frequency ultrasound(3h) showed significant difference in comparison with the CB control group (P〈0.05); vancomycin was also moderately increased with time of ultrasound to 3h. Conclusion Low-frequency ultrasonic can enhance the penetration of antibiotics through the S. epidermidis biofilm in varying degrees.
