二仙汤提取物中3种成分含量测定的高通量方法学研究 被引量:1

摘要 目的 基于96孔板多功能酶标仪,建立二仙汤提取物中总黄酮、总皂苷及总生物碱含量测定的高通量分析方法.方法 分别以淫羊藿苷、菝葜皂苷元、盐酸小檗碱为对照品,采用96孔板比色法测定二仙汤提取物中总黄酮、总皂苷、总生物碱的质量分数.结果 淫羊藿苷、菝葜皂苷元、盐酸小檗碱分别在41.12~205.6、81.12~405.6、20.34~ 61.02 μg/mL内线性关系良好,加样回收率分别为98.3%(RSD为4.44%,n=6)、101.3%(RSD为1.55%,n=6)、100.9%(RSD为2.15%,n=6).结论 96孔板比色法简单快速、通量高、准确度高、重复性好,可以用于大样本量二仙汤提取物生产过程的质量控制. Objective To establish a high-throughput method of quantitative determination for total flavonoids,total saponins and total alkaloids in Er-xian decoction based on 96-well plates Varioskan Flash multimode microplate reader. Methods 96-well plates colorimetry was used for the determination of total flavonoids, total saponins and total alkaloids in Er-xian decoction extract, with icariin, sarsasapogenine and berberine hydrochloride as reference substances, respectively. Results The colorimetry showed good specificity for quantitative analysis of total flavonoids, total saponins and total alkaloids in the extract of Er- xian decoction.The linear range of icariin, sarsasapogenin and berberine hydrochloride was 41.12-205.6 μg/ mL,81.12-405.6 μg/mL and 20.34-61.02 μg/mL, respectively.The average recoveries were 98.3% (RSD = 4. 44%, n = 6) for icariin, 101.3% ( RSD = 1.55%, n = 6) for sarsasapogenin and 100. 9% ( RSD = 2. 15% ,n= 6) for berberine hydrochloride.Conelusion 96-well plates eolorimetry was simple and rapid, high-throughput,highly accurate, reproducible, and suitable for large sample analysis in quality control of Er-xian decoction.
