摘要
目的探讨西咪替丁对体外培养的人大肠癌细胞株COLO-320的抑制和诱导其凋亡的作用。方法采用MTT比色法、形态学观察和TUNEL法检测不同浓度西咪替丁溶液处理COLO-320细胞后的生长状况、形态学、分子生物学改变。结果西咪替丁可抑制COLO-320细胞的增殖,且有时间和剂量依赖性趋势,1.0mg/L西咪替丁处理COLO-320细胞48h后,在光镜、电镜检测中可见典型的凋亡细胞;TUNEL法从分子生物学角度证实了凋亡的发生。结论西咪替丁对大肠癌细胞COLO-320有抑制作用,主要通过诱导其凋亡而发生。
Objective To investigate the effect of cimetidine on proliferation and apoptosis of hu- man colorectal cancer cell line COLO-320 in vitro. Methods Treated with different concentra- tions of cimetidine, COLO-320 cell's proliferation was determined by MTT colorimetric assay. Morphological changes of apoptotic cell were observed by light and electron microscopy. Apop- totic cells were quantitatively determined by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL). Results Cimetidine(1.0-4.0mg/L) inhibited the prolifera- tion of COLO-320 cells obviously in time and dose-dependent trends. After 48h of cimetidine (1.0mg/L) treatment, there were many morphologic features of apoptosis visible through light and electron microscope. TUNEL had confirmed the occurrence of apoptosis from the angle of molecular biology. Conclusion It is obviously that cimetidine has inhibitory effect to the COLO- 320 cells, mainly through inducing its apoptosis.
