一株新MLST型创伤弧菌的分离鉴定 被引量:4

Isolation and identification of a new Vibrio vulnificus type by MLST

摘要 目的对从住院病人脓液标本中分离的创伤弧菌(2012-DY VV1)进行病原学分析与研究。方法采用全自动微生物分析仪进行生化鉴定和耐药性分析;采用PCR方法检测溶细胞素vvhA基因,及vcg和16S rRNA基因分型;应用多位点序列分析(MLST)进行分子生物学分型。结果该菌株经生化鉴定为创伤弧菌(概率为90%);对大多数常用抗生素都敏感,但对头孢唑啉、头孢替坦中度敏感;PCR检测vvhA阳性,为vcgC-16S rRNA B基因型;菌株的序列型为glp6,gyrB 6,mdh 2,metG 12,purM 7,dtdS 15,lysA 12,pntA 7,pyrC 9,tnaA 15。结论该菌株的vcg和16S rRNA PCR分型为CB型,提示具有较强的致病能力;MLST分型为新发现的ST菌株,被命名为ST136,注册号为id-218。 Objective To study the etiological characteristics of Vibrio vulnificus strain( 2012-DY VV1) isolated from the pus sample of an inpatient. Methods The automatic microorganism analyzer was used for the biochemical identification and antimicrobial susceptibility analysis of the isolates. PCR method was performed to detect the vvhA gene and characterize the genotype of vcg and 16S rRNA loci,and multilocus sequence typing( MLST) was used for the molecular typing of the Vibrio vulnificus isolates. Results The isolate was identified as Vibrio vulnificus( 2012-DY VV1,90% probability),and sensitive to most of the commonly-used antibiotics,but moderately sensitive to cefazolin and cefotetan. The isolate was found vvhA gene positive, and identified as the vcgC-16S rRNA B genotype profile( CB). The MLST allele of 2012-DY VV1 were glp 6,gyrB 6,mdh 2,metG 12,purM 7,dtdS 15,lysA 12,pntA 7,pyrC 9,tnaA 15. Conclusion The genotype profile of 2012-DY VV1 was identified as CB type,hinting potential pathogenicity. It was confirmed as a new ST strain by the MLST method,and named as ST136 with the registration number id-218.
