MMP-2、MMP-9及其抑制因子TIMP-1、TIMP-2在宫颈癌中的表达 被引量:3

Expression and significance of matrix metalloproteinase and tissu inhibitor of matrix metalloproteinase in cervical cancer

摘要 目的分析基质金属蛋白酶2(MMP-2)和MMP-9及其抑制因子1(TIMP-1)和TIMP-2在宫颈癌不同位置中的表达情况及其临床意义。方法选取经病理证实的宫颈浸润癌患者118例(ICC组)、宫颈上皮内瘤样病变患者75例(CIN组),取病变中心组织和边缘组织;选取正常宫颈组织标本45例为对照组。采用SABC法行免疫组化检测各组中MMP-2、MMP-9、TIMP-1和TIMP-2因子表达阳性率、染色强度和表达强度。比较各组相关因子表达情况差异。结果 ICC组中MMP-2和MMP-9的阳性率、染色强度和表达强度显著高于CIN组和对照组,TIMP-1和TIMP-2的阳性率、染色强度和表达强度显著低于CIN组和对照组(P<0.05)。在ICC组,边缘癌组织的MMP-2与MMP-9的阳性率、染色强度和表达强度明显高于中心癌组织,而TIMP-1与TIMP-2的阳性率、染色强度和表达强度明显低于中心癌组织(P<0.05)。结论 MMP-2、MMP-9及其抑制因子TIMP-1、TIMP-2与宫颈癌的发生、发展密切相关,可能在宫颈癌的侵袭与转移中发挥着重要作用。 Objective To investigate the expression and significance of matrix metalloproteinase ( MMP - 2, MMP - 9) and tissu inhibitor of matrix metalloproteinase (TIMP- 2, TIMP- 1 ) in cervical cancer. Methods Totally 118 patients with invasive cervical cancer(ICC) and 75 patients with cervical intraepithelial neoplasm (CIN) were selected, and 45 ca- ses of normal cervical epithelium(NCE) were selected as control. The expression of MMP - 2, MMP - 9, TIMP - 2 and TIMP - 1 were examined by SABC method of immunnhistochemistry. Expression rate and expression level of every factor were recorded. The results were compared between the groups. Results Compared with control group and CIN group, ex- pression rate, staining intensity and expression intensity of MMP -2, MMP -9 in ICC groups were significantly higher, TIMP -2 and TIMP - I in ICC groups were significantly lower(P 〈0. 05). In ICC group, the expression rate, staining in- tensity and expression intensity of MMP - 2, MMP - 9 in the tumor edge were significantly higher than those in the tumor center, while the expression rate, staining intensity and expression intensity of TIMP -2, TIMP - 1 were significantly low- er than those in the tumor center (P 〈 0. 05). Conclusion MMP - 2, MMP - 9 and TIMP - 2, TIMP - 1 may play an important role in development, progression, invasion and metastasis of cervical cancer.
