摘要
目的构建三叶木通AFLP指纹图谱,为种质资源鉴定、品种选育奠定基础。方法采用扩增酶切片段多态性(AFLP)技术,对10份三叶木通种质进行基因组DNA多态性分析。结果从64对引物中筛选到8对高效扩增引物,获得861个多态性位点,多态性比率达到90.65%。依据多态性位点可将10个三叶木通种质聚类为三类。结论 AFLP技术适于三叶木通种质多态性分析。
Objective To establish AFLP fingerprints of Akebia trifoliata, lay the foundation for identification of germplasm re- sources,breeding. Methods The genome DNA polymorphism of 10 samples of Akebia trifoliata were detected by AFLP technique. Results Eight pairs of primers with high polymorphism and powerful distinctiveness were selected fl'om sixty - four pairs of prim- ers,and got 861 polymorphism markers. The percentage of polymorphic bands reached 90.65%. Based on the AFLP resuhs,the 10 Akebia trifoliata samples were clustered into three groups. Conclusion The AFLP reaction system is suitable for polymorphism analysis of Akebia trifoliata germplasm.
