虎杖苷调节海人酸致痫鼠颞叶、海马区P-糖蛋白表达的实验研究 被引量:5

Experimental Study of Polydatin Regulating P-Glycoprotein Expression in Temporal Lobe and Hippocampus with Kainic Acid-Induced Epilepsy Rats

摘要 目的:观察虎杖苷对海人酸致癫痫鼠颞叶、海马区P-糖蛋白表达的影响。方法:通过脑立体定向技术,将海人酸(KA)1.5μg(3μL)注射至SD大鼠海马,制作颞叶癫痫模型,假手术组为同法于大鼠海马注射3μL生理盐水。将建模成功的26只癫痫大鼠随机分为虎杖苷处理组(n=13)、生理盐水处理组即对照组(n=13)。分别给予虎杖苷(100 mg/kg)、等体积生理盐水灌胃1次/天,均连续30天。观察记录大鼠发作频率、级别及持续时间,并采用免疫组化EnVision法检测大鼠颞叶、海马区P-糖蛋白(P-gp)表达水平。结果:观察期内虎杖苷组在干预后,发作频率及级别、平均持续时间较前明显减少(均P<0.05),而对照组未见差异(均P>0.05),对照组和虎杖苷组P-gp的表达强度均高于假手术组(均P<0.05),与对照组比较,虎杖苷组P-gp的表达水平有所下降(P<0.05)。结论:虎杖苷有一定的抗癫痫作用,下调P-gp的表达可能为其机制之一,为开发新的辅助抗癫痫药物提供思路。 Objective : To observe the influence of polydatin on P - glycoprotein expression at temporal lobe, hippocam- pus in the kainic acid epileptic rats. Methods:By stereotaxic technique, the kainic acid (KA) 1.5 μg (3 μL) was injec- ted into SD rats hippocampus, making the model of temporal lobe epilepsy. 3 μL NS was injected into rats' hippocampus of the shame group in the same method. Successful model of 26 epileptic rats were randomly divided into polydatin treat- ment group (n = 13) and normal saline group (n = 13). Respectively polydatin ( 100 mg/kg) and saline were given once a day for 30 consecutive days. We observed and recorded the rat seizure frequency, level and duration. The immunohis- tochemical EnVision assay was used to measure the P - gp expression levels in the rat temporal lobe and hippocampus. Re- sults:In the observation time, polydatin treatment group, seizure frequency and level and the average duration were de- creased significantly ( P 〈 0.05 ), however, there was no difference in the control group ( P 〉 0.05 ). The P - EP expres- sion intensity of control group and polydatin treatment group was higher than that of the sham group (P 〈 0.05), and the P - gp expression level was decreased in polydatin treatment group compared with that in the control group ( P 〈 0.05 ). Conclusions : The polydatin inhibiting the expression of P - gp may be one of its mechanisms which have some antiepileptic effects, and provide ideas for the development of new auxiliary antiepileptic drugs.
