黄芪多糖对人胃癌细胞移植瘤的抑制效应及对Bcl2、Bax表达的影响 被引量:13

Inhibitory Effect of Astragalus Polysaccharides on Human Gastric Carcinoma Cells in Nude Mice and Its Impact on Expressions of Bcl2 and Bax

摘要 目的:观察黄芪多糖对人胃癌细胞SGC-7901移植瘤的抑制作用,并探讨其作用机制。方法:采用体外培养的SGC-7901人胃癌细胞制作荷瘤裸鼠模型,随机分为模型组、氟尿嘧啶组(10 mg/kg)、黄芪多糖高剂量组(1.5 g/kg)和黄芪多糖低剂量组(0.75 g/kg),每组6只,于治疗开始第0、7、14、21、28 d分别测量肿瘤体积;停药后第2 d处死裸鼠,剥离瘤组织,称取瘤重,计算其抑瘤率;取部分肿瘤组织做病理学检查;采用免疫组织化学法检测肿瘤组织中Bcl2和Bax的表达情况。结果:模型组裸鼠肿瘤体积明显增大,重量明显增加,给予相应药物干预之后,黄芪多糖高、低剂量组与氟尿嘧啶组裸鼠肿瘤增长缓慢,体积减少与重量明显减轻,与模型组相比,具有显著性差异(P<0.05,P<0.01);病理学检查结果显示,给予黄芪多糖干预后,瘤结节体积缩小,中央及边缘区域出现较大面积的坏死。免疫组化结果显示各给药组Bcl2蛋白表达明显低于模型组,而Bax蛋白表达明显高于模型组(P<0.05,P<0.01)。结论:黄芪多糖对SGC-7901细胞移植瘤具有明显的抑制作用,其机制可能与降低Bcl2的表达,增高Bax的表达,下调Bcl2/Bax比值,诱导胃癌细胞凋亡有关。 Objective:To investigate the inhibitory effects and mechanism of Astragalus olysaccharides (APS) on SGC -7901 in nude mice. Methods:Athymic mice were injected in vitro -cultured SGC -7901 human carcinoma cells and they were divided into model group,5 -FU group,APS( 1.5 g/kg) group, APS(0.75 g/kg) group. The treatment of cor- responding drugs were given for 28 days and tumor volumes were measured in 0,7^th, 14th ,21^th ,28th. At the end of experi- ment, tumor tissues were taken out and weighed. A portion of the tumor tissue was for pathological examination. The mmunohistochemistry stain was used to quantify the expression levels of Bcl2 and Bax. Result : Compared with the model group, the average tumor volume in 5 -FU group,APS( 1.5 g/kg) group and APS(0.75 g/kg) group was statistically smaller than that of the control group (P 〈 0. 05 ,P 〈 0.01 ) and the weights of tumor tissue were also reduced. Pathologi- cal examination showed that the tumor tissue was obviously improved after giving APS. Compared with the model group, the expressions of Bcl2 in 5 - FU group, APS( 1.5 g/kg) group and APS(0.75 g/kg) group were significantly lower (P 〈0.05, P〈O. 01 ) and expressions of Bax were significantly higher (P 〈0.05, P 〈0. 01 ). Conclusion:APS can signifi- cantly inhibit the growth of SGC -7901, which might be related with the down- regulation of the expression of Bcl2 and up - regulation of the expression of Bax.
