摘要
用受检者自身血浆或血清配制RPMI—1640完全培基,以外周血淋巴细胞作效应细胞,K_(562)细胞作靶细胞,~3H—TdR作指示物。用此法对19例正常人、14例治疗前鼻咽癌病人和11例治疗前原发性肝癌病人进行测定。发现治疗前肝癌患者的NK细胞功能显著低于正常人(P<0.001);治疗前鼻咽癌患者的NK细胞功能与正常人无明显差别(P>0.5)。
Patientis self serum was used in preparing the complete medium for the determination of
the human natural killer cell function. Human peripheral blood lymphcytes were used as the effector
cells. K_(562) cells of human leukemia were used as the target cells. The ratio beteen the effector cells and
the target cells was 20: 1.37×10~4Bq of ~3H--TdR was set in every hole. After fostering for 20 hours, the cells were then concentrated. The natural killer cell function was determined by measuring the 3~H--TdR strength in the target cells and calculating the cell inhibition ratio (CIR). We had examined 44 human specimens by this method. The specimens included 19 healthy controls, 14 nasopharyngeal cancer (NPC) patients and 11 primary hepatocarcinoma (PHC) patients. We found that the natural-killer cell function of the PHC patients (70. 455 ±5. 574%) was significantly lower than that of the healthy controls(80. 789± 5. 401%)(P<0. 001). But, that of the NPC patients (8O. 071± 8. 914%) did not differ significantly from that of the healthy controls ( P>0. 500).
