以EITB技术逐一分析了猪囊尾蚴两种抗原制备物中各多肽的一些生化特性和抗原性.囊液抗原至少含22种多肽,相对含量在0.1%以上的主要多肽17种,分子量11.5~118ku不等,其中11.5~26 ku的多肽为高度特异性抗原,其性质为糖蛋白,是囊虫病免疫诊断的理想抗原.29.2ku、31.5 ku、82ku、115 ku的多肽是相对特异性抗原.头节囊壁混合抗原至少含36种多肽,主要多肽28种,分子量12.7~125ku不等,其中67ku、722ku、79ku、89ku、121ku、125ku的多肽是相对特异性抗原.以11.5~26ku的多肽为诊断区带,探讨EITB在囊虫病免疫诊断中的应用价值,发现EITB的灵敏度与LISA相同,特异度则显著高于ELISA.
Some biochemical characteristics and antigenicity of the polypeptides of two antigenic prepared products of Cysticerci cellulosae were analysed step by step with EITB ech-nique.It was found that the antigens from the cyst fluid contained at least 22 polypeptides,17 of them having a relative content of more than 0.1%, and were called the major polypeptides.The molecular weights of the major polypeptides were from 11.5ku to 118ku.Of these polypeptides, the one having a molecular weight of 11.5~26ku was a highly specific antigen that could be used as an ideal antigen in the immunological diagnosis of cysticercosis.Four polypeptides (MW 29.2ku, 31.5ku, 82ku, and 115ku) were relatively specific antigens.The antigens from the scolex-cyst wall complex contained at least 36 polypeptides, of which there were 28 major polypeptides.The molecular weights of the major polypeptides were from 127ku to 125ku.Of these polypeptides, six (MW 67ku , 72.2ku ,79ku , 89ku , 121ku and 125ku) were relatively specific antigens.The polypeptide having molecular weight of 11.5~26ku was used as the diagnostic band in the evaluation of the practical value of EITB in the immunological diagnosis of cysticercosis.We found that the sensitivity of EITB equals that of ELISA, but its degree of specificity was much higher than that of ELISA.
