摘要
介绍用羊水细胞直接进行多聚酶链反应的基因分析方法.在妊娠中期,不同孕周(龄)的羊水细胞中有核的羊水细胞数为0.1×10~9/L~0.16×10~9/L.1 ml羊水细胞经处理后,可直接用于20余次PCR,不需提取DNA.在绒毛活检有困难,或孕妇情况不允许,早期产前诊断不能进行时,本法可作为简单、安全、有效的中期产前诊断方法.普通基层医院亦可取材送检,易于推广使用.
A method for gene analysis using amniotic fluid cells directly for PCR(polymerase chain reaction)was described In pregnant women of 16~27th week gestation, there were 0.1×10~9~0.16×10~9/L nucleated cells in the amniotic fluid The amniotic fluid cells were heated in sodium hydroxide solution and used directly for PCR without DNA extraction The cells from 1 ml amniotic fluid were sufficient to perform PCR for more than 20 times.This is a simple, save and reliable method for second trimesterfetal prenatal diagnosis and a complementary means when early fetal prenataldiagnosis failed.It can be applied easily even by hospitals and clinics in the rural areas.
