摘要
目的探讨三羟异黄酮对人乳腺癌细胞株MDA-MB-453(ER-,HER2-high)、MCF-7/HER2(ER+,HER2-high)和MCF-7(ER+,HER2-low)辐射敏感性的影响,并观察三羟异黄酮对HER2/neu高表达人乳腺癌细胞辐射敏感性的作用。方法通过基因转染技术建立HER2/neu高表达的人乳腺癌MCF-7细胞,免疫荧光细胞化学法鉴定;经100 mol/L三羟异黄酮联合或单独10 Gyγ射线处理MDA-MB-453、MCF-7/HER2和MCF-7三种乳腺癌细胞24 h后,采用流式细胞仪检测三羟异黄酮对乳腺癌细胞辐射敏感性的影响。结果与对照组相比,MDA-MB-453、MCF-7/HER2细胞的射线组凋亡率无显著差异(P>0.05),三羟异黄酮联合射线组的凋亡率,与射线组和对照组相比,分别增加25.51%(P<0.05)、56.16%(P<0.05);MCF-7细胞射线组凋亡率比对照组增加4.62%(P<0.05),而三羟异黄酮联合射线组与射线组比较没有显著差异(P>0.05)。结论 HER2/neu高表达的乳腺癌细胞对射线诱导的凋亡有抵抗作用,但三羟异黄酮可增强γ射线诱导HER2/neu高表达乳腺癌细胞MDA-MB-453细胞、MCF-7/HER2细胞的凋亡作用。
Objective To investigate the effects of genistein on the radiosensitivity of human breast cancer cell strains with expression of MDA-MB-453 ( ER-, HE R2-high), MCF-7 ( ER +, HER2-high), and MCF-7/HER2 ( ER +, HER2-1ow), and to observe the effects of genistein on the radiosensitivity of HER2/neu highly expressed human breast cancer cells. Methods Highly expressed HER2/neu human breast cancer cell MCF-7 was constructed through cell transfection technology and was identified by immunofluorescence cytochemical technology. After the three kinds of breast cancer cells i. e. MDA-MB-453, MCF-7/HER2, and MCF- 7 were treated by 100 μmol/L genistein combined with or simply use of 10 Gy γ ray,flow cytometry was used to analyze the effects of genistein on the radiosensitivity of the breast cancer cells. Results There was no significant difference between the apoptotic rates of MCF-7/HER2 and MDA-MB-453 cells treated by γ ray compared with those of the control group ( P 〉 0.05 ). The apoptotic rates of those cells treated by genistein combined with γ ray increased by 25.51% ( P 〈 0.05 ) and 56.16% ( P 〈 0.05 ), respectively, compared with those of the cells treated only by γ ray and the control group. The apoptotic rate of MCF-7 cells treated by γ, ray increased by 4.62% (P 〈0.05)compared with that of the control group(P 〈 0. 05),but there was no significant difference between the apoptotic rates of the cells in the γ ray and the control group ( P 〉 0.05 ). Conclusions HER2/neu highly expressed breast cancer cells have resistant effect on the apoptosis induced by γ ray,but genistein can increase the apoptotic effect of HER2/neu highly expressed human breast cancer cells induced by γ ray.
