摘要
探讨了用野生油菜黄单孢菌发酵生产黄原胶的发酵条件及其提纯分离的技术。黄原胶发酵的最佳温度为 2 8~30℃ ,接种量为 1 0 % ,蔗糖作为碳源优于葡萄糖。当搅拌速度为 50 0r/min,空气通入量为 0 5min- 1 (跟反应器的体积比 ) ,产胶阶段的pH值 7 0。采用酶降解和硅藻土吸附提纯 ,用超滤技术浓缩提纯后的发酵液 ,然后用乙醇析出浓缩液中的黄原胶。
The fermentation conditions of Xanthomonas Campestris and separation technique of xanthan gum was introduced.The optimal fermentation conditions are:temperature 28~30℃,the inoculation quantity of Xanthomonas Campestris 10%,sucrose concentration 5 g/L,agitation rate 500 r/min,air input rate 0 5 min -1 (volume space velocity),and pH nearby 7 0.The insoluble microbe was degraded by enzymes and insoluble solid absorbed by diatomite.Then ultra filtration was used to concentrate the treated broth,and ethanol to precipitate xanthan gum.
