摘要
目的多囊卵巢综合征(polycystic ovary syndrome,PCOS)与卵巢类固醇的过度产生、促黄体生成素的分泌增加以及胰岛素抵抗等相关。为了进一步研究肥胖与PCOS发病的关系,文中通过建立雌性大鼠的高脂肪饮食模型,来探索高脂肪饮食的大鼠是否具有PCOS的表型。方法选SD雌性大鼠,喂以高脂饮食17周,建立大鼠的高脂肪饮食模型(高脂组),并与普通饲料喂养大鼠组(对照组)比较。结果高脂肪饮食17周后,与对照组比,高脂组空腹血糖升高[(7.02±0.48)mmol/L vs(6.00±0.70)mmol/L,P<0.01],总胆固醇[(2.04±0.15)mmol/L vs(1.70±0.42)mmol/L,P<0.05]和低密度脂蛋白亦升高[(0.24±0.07)mmol/L vs(0.15±0.04)mmol/L,P<0.01],大鼠出现胰岛素抵抗和高脂血症表现,卵巢形态学呈多囊状改变,体内雄激素水平也有所升高。结论高脂肪饮食条件可以诱导雌性大鼠发生PCOS的表型改变,提示高脂饮食可能是女性多囊卵巢的发病诱因之一。
Objective Exaggerated ovarian steroid production, increased secretion of luteinizing hormone and insulin resist- ance are concerned with polycystic ovary syndrome (PCOS). The aim of this study is to further study the correlation of obesity and PCOS, the high-fat diet model of female rats was established to explore whether the high-fat diet rats have phenotype of PCOS or not. Methods The female rats were fed with high-fat diet for 17 weeks to set up the high-fat diet model(high-fat group) which was com-pared with the control group fed with normal diet. Results After 17 weeks of high-fat diet, the fasting blood-glucose of high-fat group rose compared with control group ( [ 7.02 _± 0.48 ] mmol/L vs [ 6.00 ± 0.70 ] mmol/L) ( P 〈 0.01 ). The total cholesterol ( [ 2.04 ± 0.15 ] mmol/L vs [ 1.70 ± 0.42] mmol/L) ( P 〈 0.05 ) and low density lipoprotein ( [ 0.24 ± 0.07 ] mmol/L vs [ 0. 15 ± 0.04]mmol/L) (P 〈0.01 ) rose at the same time. The female rats showed insulin resistance and hyperlipidemia. The ovaries ap- peared the cystic follicles and the androgen levels increased. Conclusion High-fat diet could cause the appearance of cystic follicles in female rats' ovaries. These results suggest that high-fat diet is one of the inducements of PCOS.
