从LOC440040、SYCP2L等mRNA角度探讨慢性乙型肝炎脾胃湿热证的客观化研究

摘要 【目的】从LOC440040、SYCP2L、MYH6、MDGA1、C17orf97、LIPC、ATOH8等mRNA角度确立慢性乙型肝炎(简称"慢乙肝")脾胃湿热证客观化辨证依据。【方法】研究对象分为2组:慢乙肝无症状者组、慢乙肝脾胃湿热证组,借助荧光定量PCR技术检测患者静脉血LOC440040、SYCP2L、MYH6、MDGA1、C17orf97、LIPC、ATOH8等mRNA水平。【结果】慢乙肝脾胃湿热证患者C17orf97、LIPC、ATOH8的mRNA表达水平显著降低,差异有统计学意义(P<0.05或P<0.01),而慢乙肝脾胃湿热证患者LOC440040、MDGA1、SYCP2L、MYH6的mRNA表达水平显著升高,差异有统计学意义(P<0.01)。【结论】LOC440040、SYCP2L、MYH6、MDGA1、C17orf97、LIPC、ATOH8等差异基因谱可以作为慢乙肝脾胃湿热证客观化的诊断标志物之一。 Objective To find the obiectified hepatitis B (CHB) by detection of mRNA markers for spleen-stomach damp-heat syndrome of expression levels of differentially expressed genes LOC440040, SYCP2L, MYH6, MDGA1, C170orf97, LIPC, ATOH8, etc. with polymerase chain chronic such as reaction(PCR) methods. Methods The subjects were divided into two groups, namely the group of CHB patients without symptoms, and the group of CHB patients with spleen-stomach damp-heat syndrome.Fluorescent quantitation PCR was adopted for the detection of mRNA expression levels of LOC440040, SYCP2L, MYH6, MDGA1, C17orf97, LIPC, ATOH8 in venous blood. Results In comparison with CHB patients without symptoms, CHB patients with spleen-stomach damp-heat syndrome had low C17orf97, LIPC, ATOH8 mRNA expression levels (P 〈0.05 or P 〈0.01), and had high LOC440040, MDGA1, SYCP2L, MYH6 mRNA expression levels (P 〈0.01) . Conclusion The mRNA expression levels of differentially expressed genes of LOC440040, SYCP2L, MYH6, MDGA1, C17orf97, LIPC, ATOHS, etc. can be used as one of the objectified markers for spleen-stomach damp-heat syndrome of CHB.
