摘要
由海产贝类中分离到一菌株 YS- 94 12 - 130 ,为革兰氏阴性短杆菌、末端圆、有鞭毛、不能滑行运动、大小为 0 .4~ 0 .7μm× 1.0~ 1.5μm,裂殖生殖 ,不生成芽孢 ,胞内无多 -β-羟基丁酸颗粒。菌落光滑 ,黄色、半透明 ,边缘完整。其色素无荧光 ,含类胡萝卜素 ,其正己烷提取液经光谱测定最大吸收峰为4 50 nm。该菌生长的温度范围为 0~ 2 5℃ ,最适为 18~ 2 2℃ ,最高 2 5℃ (在 2 5℃生长极弱或几乎不生长 )。此菌株于 p H7.0~ 9.0能生长 ,最适 p H为 7.5,耐 6% Na Cl,80μg/ m1的链霉素或 80μg/ ml的氨卞青霉素。 YS- 94 12 - 130菌株为好气菌 ,严格的有氧呼吸代谢 ,过氧化氢酶、氧化酶阳性和磷酸酶阳性 ,利用多种糖、水解淀粉但不分解纤维素、琼脂和几丁质。水解干酪素、胨化石蕊牛奶最后变碱。液化明胶试验阴性 ,还原硝酸盐 ;不产 H2 S和吲哚 ;MR和 V.P.试验阴性 ;YS- 94 12 - 130菌株 DNA中 G+C含量为 32 .2 mol%。该菌株的特征符合黄杆菌属的特征 ,但与此属中的其他种比较又有显著的差别 ,因此将其定为黄杆菌属一新种 ,命名为黄海黄杆菌 ( Flavobacterium yellowsea sp.nov)。
The strain YS 9412 130 is isolated from marine shells. The cells of strain are short rods with rounded ends, measuring 0 4~0 7μm by 1 0~1 5μm, gram positive, flagellated, not gliding movement, reproduction by schizogenesis. Endospores not formed. Intracellular granules of poly β hydroxybutyrate are absent. Colonies of strain YS 9412 130 are yellow colour, translucent and smooth with entire edges. The pigments are non fluorescent, and include carotenoids. The maximum absorption peak of the pigments in n hexane is 450nm.The temperature range for growth is 0 to 25℃, minimum, 5℃, optimum, 18 to 22℃; maximum, 25℃(very week growth or almost no growth ); the pH range for growth is 7 0~9 0, optimum growth occurred at pH7 5. This strain may be tolerant of 6%NaCl, 80μg/ml of streptomycin and of ampicillin. Strain YS 9412 130 is aerobic, having a strictly respiratory type of metabolism. Catalase ,oxidase and phosphatase positive. Many sugars are utilized and alkalified. It hydrolyzes starch, but cellulose (CMC or filter paper), chitin and agar are not digested. This strain hydrolyzes casein, and peptonizes litmus milk and turns it into alkaline. Gelatin isnt liquefied. Nitrate is reduced to nitrite. M.R. and V.P. tests are negative. H 2S and indole are not produced. The guanine plus cytosine content of deoxyribonucleic acid of the strain YS 9412 130 is 32 2mol%. This strain belongs to the genus Flavobacterium. When it is compared with descriptions of Flavobacterium ,it differs significantly from strains of previously described species. Therefore, it is regarded as a new species. According to its character of growth, the name Flavobacterium yellowsea sp.nov is proposed.
