摘要
目的评价灭活轮状病毒(rotavirus,RV)与灭活肠道病毒71型(enterovirus71,EV71)联合疫苗的免疫效果及两种抗原间的相互作用。方法将灭活RV与EV71按不同抗原含量配比制成联合疫苗免疫ICR小鼠,同时设单价疫苗组,均经小鼠腿部肌肉注射,隔2周免疫1次,共2次,分别于每次免疫后2周,经小鼠尾静脉采血,分离血清,经56。c灭活30rain后,采用血清中和试验检测小鼠血清中抗RV和抗EV71中和抗体效价。结果初次免疫后14d,各组小鼠血清中RV特异性中和抗体阳转率为50%-67%,中和抗体效价为2.2-2.8,加强免疫后14d,抗体阳转率达100%,中和抗体效价为28.5-35.9。初次免疫后14d,各组小鼠血清中抗EV71中和抗体阳转率为83%-100%,中和抗体效价为4.0-7.1,加强免疫后14d,中和抗体阳转率达100%,中和抗体效价为25.4-71.8;联合疫苗组与单价疫苗组间抗RV及抗EV71中和抗体效价差异无统计学意义(P〉0.05);Rv和EV71抗原量按160/160EU配比时,可诱导出最高的抗RV抗体及抗EV71抗体应答。结论Rv与EV71联合疫苗可诱导小鼠产生针对两种病毒的体液免疫应答,抗体应答水平与疫苗抗原配比剂量相关,未发现两种抗原间有协同或拈抗作用。
Objective To evaluate the immune effect of inactivated rotavirus (RV) and enterovirus 71 (EV71) combined vaccine and the interaction between the two antigens. Methods Combined vaccine was prepared with inactivated RV and EV7l at various ratios and immunized i.m. to ICR mice for 2 times at an interval of 2 weeks, using the corresponding monovalent vaccines as control. Serum samples were collected 2 weeks after each immunization, inactivated at 56 °C for 30 min, and determined for neutralizing antibody titers against RV and EV71 by serum neutralization test. Results The positive rates of neutralizing antibody against RV in sera of mice in various groups 14 d after the first immunization were 50% N 67%, while the titers were 2. 2 N 2. 8. However, the positive rates reached 100% 14 d after the second immu- nization, while the titers were 28.5 - 35.9. The positive rates and titers of neutralizing antibody against EV71 14 d after the first immunization were 83% - 100% and 4.0 - 7. 1, while those 14 d after the second immunization were 100% and 25.4 - 71. 8, respectively. The neutralizing antibody titers against RV and EVTI induced by combined vaccine showed no significant difference with those by the corresponding monovalent vaccine (P 〉 0. 05 ). The RV and EV71 antigens at a ratio of 160/160 EU induced RV and EV71 antibody responses at satisfactory levels. Conclusion RV and EV71 combined vaccine induced humoral immune response against the two viruses in mice. The response level was associated with the ratio of the two antigens. However, no synergistic or antagonistic effect between EVTI and RV antigens were observed.
