摘要
目的评价丹参酮ⅡA磺酸钠对2型糖尿病大鼠心肌肿瘤坏死因子(TNF)-αmRNA表达的影响,探讨其对糖尿病心肌病的保护机制。方法将大鼠随机分为正常组、模型对照组及丹参酮ⅡA磺酸钠组。成模后丹参酮ⅡA磺酸钠组给药12周,留取血及心肌标本用放免法检测各组大鼠血浆TNF-α,RT-PCR方法检测心肌组织中NF-κB、TNF-αmRNA表达水平。结果模型对照组大鼠血浆中TNF-α水平较正常对照组明显升高(P<0.01),且心肌组织中NF-κB、TNF-αmRNA表达较正常对照组明显增加(P<0.01)。给药12周后,丹参酮ⅡA磺酸钠组心肌组织中NF-κB、TNF-αmRNA表达较模型对照组明显降低,两组之间差异显著(P<0.05)。结论丹参酮ⅡA磺酸钠可能通过抑制2型糖尿病大鼠心肌组织中NF-κB通路,减少TNF-αmRNA表达,对2型糖尿病大鼠心肌提供保护作用。
Objective:To evaluate the effect of tanshinone Ⅱ A sulfonate sodium on TNF-α mRNA expressions in myocardium of rats with type-Ⅱ diabetics and further discuss the protective mechanism of diabetic cardiomyopathy.Methods:Wister rats were randomly divided into normal control group (NC),model control group (DM) and tanshinone Ⅱ A sulfonate sodium treatment group (DMS).After rats model was established and STS was administrated for 12 weeks.Specimens of blood and myocardium in rats were reserved for detecting serum TNF-α by radio immunoassay and expressions of NF-κB (Nuclear factor-kappa B) 、TNF-α mRNA in myocardium by RT-PCR.Results:Compared with the NC group,the level of TNF-α in DM group elevated (P< 0.01).Compared with the NC group,the NF-κB and TNF-α mRNA expressions in myocardium of DM group significantly increased (P< 0.01).However,the NF-κB and TNF-α mRNA expressions in myocardium of DMS group after 12 weeks treatment were less than those of DM group,showing statistical differences (P< 0.05).Conclusion:Tanshinone Ⅱ A sulfonate sodium can reduce TNF-α mRNA expressions in myocardium of rats with type-Ⅱ diabetics by inhibition of NF-κB pathway and protect the diabetic rats' myocardium.
