DNA梯度化分析软件系统在细胞凋亡定量检测中的应用

摘要 目的:检验我们设计的DNA梯度化分析软件在细胞凋亡定量检测中的应用价值 方法:用Camptothesin(CAM)诱导U937细胞株凋亡,通过该系统计算DNA琼脂糖凝胶电泳后DNA的梯度值,并与凋亡细胞胞浆寡聚核小体的Elisa检测值进行相关性分析.结果:随着CAM浓度的增加,DNA琼脂糖凝胶电泳后梯度化程度越明显,测出的梯度值也越大,而且与胞浆寡聚核小体的Elisa检测值呈正相关(r=0.989,p<0.01)结论:利用DNA梯度化分析系统可对琼脂糖凝胶电泳后的凋亡细胞DNA片段,进行定量分析,提供简便直接的方法. Porpose:To assess the application value of DNA laddering analysis system designed by us in quantitative measurement of programmed cell death. Methods: Camptothecin was used to induce apoptosis of U937 cells, DNA agarose gel electrophoresis was performed and DNA laddering value was measured by DNA laddering analysis system, which was then correlated to the value measured by cellular DNA fragmentation Elisa. Results: With the increase of CAM concentration, laddering pattern was much more obvious in DNA agarose gel electrophoresis , and DNA laddering value measured by the system increased. DNA laddering value was positively correlative to the OD value measured by cellular DNA fragmentation Elisa(r=0.989,p<0.01) . Conclusion:DNA laddering analysis system could be used to measure the degree of DNA laddering in electrophoresis. Thus it can be used as a simple and direct method to quantitatively measure apoptosis.
