摘要
目的观察益气通络丹 (YQTLD)含药血清对培养SD大鼠胸主动脉平滑肌细胞 (VSMC)的影响 ,探讨该药抗冠心病、脑梗塞等心脑血管疾病的作用机制。方法血清药理学方法制备含药血清 ;MTT法测定含药血清对VSMC增殖活性的影响 ;NO试剂盒测定含药血清对VSMC作用与NO的关系。结果YQTLD含药血清能显著降低培养的SD大鼠VSMCMTT法的D(()值 ,与空白血清组有显著性差异 (P <0 0 1) ;YQTLD含药血清组培养基中NO含量明显增多 ,与空白血清组有显著性差异 (P <0 0 1)。结论YQTLD有抗VSMC增殖的作用 ,此作用可能与NO有关 ,YQTLD可能通过抗VSMC增殖 ,促进NO释放来延缓AS的发生、发展 ,从而在冠心病、脑梗死等心脑血管疾病中发挥作用。
Objective To observe the effect of serum containing YQTLD on SD rat cultured VSMC Method Serum containing YQTLD was prepared by serum pharmacological method Cultured VSMC was treated with the serum, and the influence of YQTLD on proliferating activity of VSMC was determined by MTT test The amount of NO was determined by NO test kit Results (1) Serum containing YQTLD could decrease the OD value of VSMC tested by MTT method, and there was significant difference (P<0 05) compared with the control serum; (2)The amount of NO in the culture medium of serum containing YQTLD which could inhibit the proliferation of VSMC increased remarkably, and there was significant difference (P<0 05) compared with the control serum Conclusion (1) YQTLD could inhibit the proliferation of VSMC (2) The mechanism might relate to the increase of NO in the culture medium (3) The mechanism of YQTLD's effect on vascular disease, such as coronary heart disease, cerebral thrombosis might be associated with its inhibit effect on VSMC proliferation
