基因重组人睫状神经营养因子单克隆抗体的制备及初步鉴定

摘要 目的 制备基因重组人睫状神经营养因子 (rhCNTF)单克隆抗体。方法利用rhCNTF免疫BALB/C小鼠 ,取其脾细胞与小鼠NS -1骨髓瘤细胞融合 ,制备 3株抗rhCNTF的单克隆抗体 ,分别命名为CA2 、CA6 和BB11。结果 3株抗体类型均为IgG2a,免疫印迹试验证明为特异性抗CNTF的单克隆抗体 ;抗体相加试验表明 3株单抗是针对CNTF两个不同的抗原决定簇。 Objective To produce monoelonal antibodies against recombinant human ciliary neurotrophic factor (rhCNTF) Methods rhCNTF was used to immune BALB/C mouse, the spleen cells from which were fused with murine NS-1 myeloma cells 3 monoclonal antibodies against rhCNTF were produced from the hybridoma, named CA2, CA6 and BB 11 respectively Results All the 3 antibodies were in the IgG2a subclass and proved to be specific antibodies against CNTF By Western blot experiment, McAb additivity test showed that the 3 antibodies recognize 2 different epitopes
