摘要
目的 建立一种电压钳位技术检测细胞内钙振荡的方法。方法 采用全细胞电压钳位法检测非洲爪蟾卵母细胞微量注射CaCl2 诱发的振荡电流。结果 振荡电流持续 1h以上 ,并分别被注射EGTA和肝素以及细胞外灌流氮氟灭酸所取消。结论 结果提示该振荡电流为钙诱发的振荡电流。以同样的方法也能测得氨甲酰胆碱激动表达于非洲爪蟾卵母细胞膜之大鼠脑胆碱受体而产生的振荡电流。
Objective To eastablish a measurement of Ca 2+ oscillation in cell by voltage clamp technique Method The currents induced by microinjection of CaCl 2 were measured with whole cell voltage clamp technique in Xenopus oocytes Result The currents had over 1 h duration and were blocked by microinjection of EGTA, heparin and perfusion of Niflumic acid, respectively Conclusion The results suggest that the oscillation currents were induced by CaCl 2, and the current produced by flowing carbamylcholine in the oocyte injected with rat brain mRNA previously can also be measured with voltage clamp technique
