摘要
目的 :建立测定血浆样品中西沙必利浓度的HPLC法。方法 :色谱条件 :以InertsilODS 3 ( 5 μm ,4 .6× 2 5 0mm)为色谱柱 ;乙腈 0 .0 67mol·L-1pH 4 .7磷酸盐缓冲液 ( 3 8∶62 )为流动相 ,检测波长 2 76nm ;采用固相小柱法萃取。结果 :3种浓度 3 ,5 0 ,10 0ng·ml-1萃取回收率分别为 83 .5 0 % ,90 .66% ,89.4 2 % ;日内、日间RSD分别为 3 .2 1% ,2 .74 % ,1.0 2 %及 3 .77% ,3 .74 % ,2 .5 2 % (n =4 )。血浆中药物最低检测浓度为 1ng·ml-1,线性范围 3～ 10 0ng·ml-1,r=0 .9997。结论 :方法准确 ,灵敏 ,简便。
OBJECTIVE:To determine plasma concentration of cisapride. METHODS:An Inertsil ODS-3 column was used, the mobile phase consisted of acetonitrile- 0.067 mol·L -1 pH 4.7 phosphate buffer(38∶62),the detection wavelength was 276 nm. RESULTS:The linear range was 3～100 ng·ml -1 ,r= 0.9997 .The recovery rate of high,middle ,low concentration were 89.42 %, 90.66 %, 83.50 % (n=4),coefficients of inter-day and intra-day variations were 1.02% , 2.74% , 3.21% and 2.52% , 3.74% , 3.77% (n=4). CONCLUSIONS:This method is rapid ,accurate.
