高效液相色谱法测定两性霉素B在家兔体内的血药浓度及药动学 被引量:5

摘要 目的 :建立血浆中两性霉素B浓度的HPLC测定方法 ,并用此方法研究两性霉素B在家兔体内的药动学。方法 :血浆经处理后 ,采用PlatinumC18柱以乙腈 0 .0 0 5mol·L-1EDTA 2Na( 60∶4 0 )为流动相 ,在紫外 4 0 5nm处检测。结果 :两性霉素B浓度在 0 .0 5~ 12 .8μg·ml-1范围内与色谱峰面积呈良好的线性关系 ,血浆中药物最低检测浓度为 0 .0 0 5 μg·ml-1;高中低三种浓度的日内及日间RSD % (n =5 )分别为 ( 0 .3 4~ 1.71) %和 ( 0 .2 5~ 6.78) % ;方法回收率和绝对回收率分别为 ( 10 0 .0 3~10 5 .19) %和 ( 92 .18~ 95 .2 1) % ;药动学研究表明 ,家兔静脉注射单剂量的两性霉素B后其体内血药浓度时间数据符合权重为1/C的三室模型。结论 :本方法操作简便 ,结果准确可靠 ,能很好的满足两性霉素B血药浓度测定及药动学的研究。 OBJECTIVE:To establish an HPLC method for the determination of amphotericin B (AmB) in rabbits plasma and investigate the pharmacokinetic characteristics of AmB by intravenous administration. METHODS:Platinum C 18 column and acetonitrile-EDTA-2Na(60∶40) as mobile phase, with UV detection at 405 nm.RESULTS:A good linear relationship was obtained between the peak area and the concentration of AmB from 0.05 ~ 12.8 μg·ml -1 , and the limit of detection was 0.005 μg·ml -1 . The RSD of intra-day and inter-day variations were 0.34 %~ 1.71% and 0.25 %~ 6.78 % respectively. The absolute and relative recoveries were 92.18 %~ 95.21 % and 100.03 %~ 105.19 % respectively. The concentration-time data of AmB after intravenous administration were fitted by three compartments with weight of 1/C. CONCLUSIONS:A simple, highly sensitive and selective method for the determination of AmB level in plasma was established,which could be used for its pharmacokinetic studies.
