摘要
顺铂是目前临床上最常用也是最有前途的抗癌药物之一,特别是应用在睾丸癌和乳腺癌上。肿瘤组织及肾组织分别是顺铂抗癌作用及毒性作用的靶组织;血浆超滤液中铂则代表了血液中(氵申)蛋白结合的活性铂。为了提高药物治疗疗效和控制药物毒性反应,有必要了解顺铂在体内的处置及合并用药后顺铂在体内处置的变化,因此需要建立生物组织及超滤液中铂浓度的测定方法。
Flameless atomic absorption spectrophotometry (FAAS) is adapted to the quantitative of platinum in biological tissues and fluid. Tissue Sample is digested with digestive pot, plasma ultrafiltrate is mixed by 0.25% TritonX-100. With these procedure, platinum in samples can be determined sensitively and accurately. The limit of detection is 5ppb, The recovery and the relative standard derivation are 92-99% and 3-7%. The methodology can be applied to the measurement of the distribution and disposition of the anticacer drug cisplatin.
