反向、巢式、降落PCR技术克隆奥尼罗非鱼MSTN基因3'侧翼序列 被引量:1

摘要 为获得奥尼罗非鱼侧翼MSTN基因的3'未知序列以完成奥尼罗非鱼MSTN基因全序列的克隆,采用反向、巢式、降落等3种PCR技术对该未知序列进行克隆,对克隆到的序列进行测序,并与已有的奥尼罗非鱼MSTN基因核苷酸序列进行比对,获得MSTN基因3'端1 376 bp新序列,从而完成奥尼罗非鱼MSTN基因全序列的克隆,为奥尼罗非鱼MSTN基因功能的进一步研究以及奥尼罗非鱼的分子育种研究奠定基础。说明反向、巢式、降落PCR相结合是扩增已知基因侧翼序列行之有效的方法。
