摘要
果汁量 4%的TiO2 作为吸附剂 ,能够吸附菠萝榨汁中 96 %的果菠萝蛋白酶活力 ,然后以 0 4M pH6 6的柠檬酸缓冲液可有效地进行洗脱 ,洗脱液先用截留值 2万的超滤膜超滤 ,流出液再用截留值为 5万的超滤膜超滤 ,最后所得截留液中果菠萝蛋白酶比活力达 91 1单位 /毫克蛋白 ,活力回收率为 5 4% ,纯化倍数 7 9,蛋白质溶液浓缩 1
of fruit bromelain activity is absorbed by 4% of TiO 2(by weight).It can be eluted by 0.4M citrate buffer solution(pH6.6).Ultrafiltration is applied to the elution with a membrane of 2000MWCO,and the outlet is collected and re-filtrated by a membrane of 50000MWCO.The specific activity of the remaining solution is 911Unit/mg protein,the activity recovery ratio is 54%, purification rate is 7.9. The solution is condensed in the meantime.
