摘要
目的 运用高效液相-高分辨飞行时间质谱(HPLC-TOF/MS)技术快速鉴别中药猫人参化学成分。方法 色谱分离采用SHISEIDO MG C18(3.0 mm×100 mm,3μm)色谱柱;流动相为乙腈和0.1%甲酸水溶液,梯度洗脱,0~10 min:10%~25%A,10~35 min:25%~90%A,35~40 min:90%A;柱温25℃;流速0.6 mL.min-1,柱后分流比为2:1。质谱定性采用飞行时间质谱,电喷雾离子源(ESI),正离子模式,质量数扫描范围m/z 100~1 000。结果 一次性鉴别出猫人参中15种的化学成分,其中2种成分首次从猫人参中发现。结论 建立了一种基于HPLC-TOF/MS技术对猫人参中化学成分进行快速鉴别的方法,为中药猫人参的质量控制及体内的深入研究奠定了基础。
Objective To analyze the chemical constituents in Actinidia valvata Dunn. by high-performance liquid chromatography-time of flight mass spectrometry(HPLC-TOF/MS). Methods The separation was performed on a SHISEIDO MG C18 reverse phase column(3.0 mm×100 mm, 3 μm). The mobile phase consisted of water(containing 0.1% formic acid) and acetonitrile was used as gradient elution: 0- 10 min: 10%- 25%A; 10- 35 min: 25%- 90%A; 35- 40 min: 90%A. The flow rate was 0.6 mL·min- 1, the post-column split ratio was 2 :1, and the temperature of column was 25 ℃. Time-of-flight mass spectrometer(TOF/MS) and electrospray ion source(ESI) were applied in qualitative analysis under positive ion mode, and mass scan range was m/z 100- 1 000. Results A total of 15 of the major chemical constituents in Actinidia valvata Dunn., including 2 new compounds, were identified by HPLC-TOF/MS. Conclusion A rapid and efficient HPLC-TOF/MS method to determine the chemical constituents in Actinidia valvata Dunn. has been established, which paves a way for the quality control and further studies of the herb in vivo.
