应用PFGE检测技术进行一起洋葱伯克霍尔德菌感染溯源 被引量:4

Application of the PFGE technique in tracing the source of Burkholderia cepacia infection

摘要 目的探讨利用脉冲场凝胶电泳(Pulsed field gel electrophoresis,PFGE)技术对洋葱伯克霍尔德菌引起的医院感染进行调查及溯源检测。方法对医院感染病人无菌手术伤口及治疗过程中使用的辅料、器材、器械等样本进行细菌培养,将检出的洋葱伯克霍尔德菌采用PFGE技术进行分子分型,利用BioNumerics软件对PFGE分型图谱进行数据处理,绘出聚类分析图。结果医院感染病人无菌手术伤口样本、B超探头样本、三维彩超探头样本、已启用的医用超声耦合剂样本、未启用的医用超声耦合剂样本均检出洋葱伯克霍尔德菌,所有检出菌株的PFGE图谱一致,聚类分析为同一来源,相似度为100%,据此查明感染的源头。结论脉冲场凝胶电泳(PFGE)技术应用于洋葱伯克霍尔德菌引起的医院感染暴发调查及追踪溯源,是一种快速、可靠、准确的实验室调查方法。 Objective Using pulsed field gel electrophoresis technology (short for PFGE) to investigate and trace the source of nosocomial infection due to Burkholderia cepacia. Methods Firstly, sterile surgical wound of patients with hospital infection, accessories, equipment, instruments and others used in the course of treatment were carried out bacterial culture. The burkhold- eria cepacia found by bacterial culture were detected with PFGE molecular typing methods, and the Bionumerics software was used for data processing of PFGE typing diagram, and drawing diagram for clustering analysis. Results The burkholderia cepa- cia were detected from the samples of sterile surgical wound of patients with hospital infection, B - mode ultrasound probe, the three - dimensional color doppler ultrasound probe, enabled ultrasonic coupling agent and unused ultrasonic coupling agent. All the strains showed the same PFGE diagrams, and the clustering analysis proved they were from the same source, and the simi- larity was 100%. Conclusion For examination in laboratory, pulsed field gel electrophoresis is an accurate, reliable and rapid method for investigating and tracing the source of nosocomial infection by burkholderia cepacia.
