The susceptibility of culture cells to severe fever with thrombocytopenia syndrome virus

摘要 目的 了解发热伴血小板减少综合征布尼亚病毒(SFTSV)细胞嗜性,加强对SFTSV传播与致病机制的理解.方法 通过不同感染复数(multiplicity of infection)的SFTSV感染Vero等10种不同组织来源的细胞株,采用双抗体夹心ELISA法和间接免疫荧光实验(IFA)检测不同时间点的细胞培养上清和细胞内病毒抗原表达水平,绘制病毒动态生长曲线,并观察记录细胞病变(cytopathic effect,CPE)情况,确定SFTSV细胞嗜性.结果 Vero、HEK 293、COS-7、CHO、HepG2、Hela、THP-1和MRC-5等不同组织来源的细胞均可感染SFTSV,并产生明显细胞病变,但病毒在不同细胞中的复制水平差异显著,其中Vero细胞对SFTSV较易感,病毒可增殖至10S0TCID50/ml以上;Raji细胞、Jurkat细胞不能感染SFTSV.结论 SFTSV具有广嗜性,可以感染肝、肺、肾、子宫和卵巢等多器官以及免疫系统等来源的细胞系,但不能感染T和B淋巴细胞源细胞系,为SFTSV相关基础研究提供重要线索. Objective To learn the susceptibility of culture cells to Severe fever with thrombocytopenia syndrome virus (SFTSV) and strengthen the understanding for transmission and pathogenic mechanism of SFTSV.Method SFTSV infected different cells which derived from 10 cell lines such asVero cells by different multiplicity of infection (MOI).The titration of cell culture supernatant at different time points and the expression of virus antigen were measured by double antibody sandwich ELISA and indirect immunofluorescence assay (IFA).By drawing dynamic growth curve of SFTSV and observing cytopathic effect (CPE) to determine the cell tropism of SFTSV.Result The results suggested that SFTSV can affect Vero,HEK 293,COS-7,CHO,HepG2,Hela,THP-1 and MRC-5,and significant CPE can be observed,but the virus replication levels are significantly different in these cells.Vero cells can relatively susceptible to SFTSV,and the virus titration can reach 1080 TCID50/ml.SFTSV cannot susceptibly affect Raji cells and Jurkat cells.Conclusion SFTSV has broad tissue and cell tropism,and it can infect liver,lung,kidney,uterus,ovaries and other organs as well as the cell lines of immune system,but not T and B lymphocyte-derived cell lines.The study provides important clues to the basic research of SFTSV.
