一株H5N2亚型流感病毒的全基因组序列分析及致病性的研究

摘要 本研究对2012年从我国南方的鸭群环境水样品中分离到的一株低致病性H5N2亚型流感病毒EN/HuN/S3794/2012(H5N2)进行全基因组序列和进化分析、抗原性差异分析及致病性试验。序列分析显示:其HA片段裂解位点无多个连续碱性氨基酸,具有低致病性流感病毒的特征。该病毒基因组序列与A/DK/HuN /S4120/2011 (H5N2)全基因组核苷酸序列高度同源,同源性在97.4 %~99.3 %之间,抗原性分析结果表明,该分离株与H5亚型流感病毒参考株抗原性差异较大。以106 EID50/100 μL剂量鼻腔感染4周龄SPF鸡及SPF鸭,病毒滴定未检测到病毒存在,表明该病毒不能在SPF鸡和SPF鸭体内有效复制;以106 EID50/50 μL剂量鼻腔感染6周龄BALB/c小鼠,脏器滴定结果显示,仅在肺脏中检测到病毒,小鼠体重无明显变化,表明该病毒分离株对小鼠感染能力弱,呈低致病性。 A H5N2 subtype influenza virus which isolated from southern China ducks environment was studied on sequence and genetic evolution analysis, antigenic differences analysis and animal test. Sequence analysis results showed that the HA cleavage site did not contain multiple consecutive basic amino acid, which had the characteristic of low pathogenic influenza virus. The whole genome sequence nucleotide homology between EN/HuN/S3794/2012 (H5N2) and DK/HuN/S4120/2011 (H5N2) were 97.4% and 99.3%. In addition, the virus isolate was unable to effectively replicate in four week-old SPF chickens and ducks when inoculated intranasally at the dose of 106 EID50/100 μL. And the virus existed only in lungs of six week-old BALB/c mice when infected with 106 EID50/50 μL of virus isolate, but weight gain of mice did not change significantly, indicating the infectious ability of the isolate was a low-pathogenic virus.
