摘要
目的探讨不同染色时间和试剂浓度对细胞形态测量结果的影响情况。方法以瑞氏-姬姆萨染色的红细胞为研究对象,采用不同染色时间(10、20、40 min)和A、B液比例(1∶1、1∶2、1∶4)对血涂片进行染色,应用图像分析软件分别测量红细胞的红、绿、蓝三基色和面积、平均直径、周长,并对测量结果进行分析比较。结果不同染色时间和试剂浓度组红细胞红、绿、蓝三基色差异均有统计学意义(P<0.05),其中红、绿、蓝基色值在短时间(10 min)和低浓度(1∶4)组最小,在长时间(40 min)和高浓度(1∶1)组最大(P<0.05)。不同染色时间和试剂浓度组红细胞的面积、平均直径、周长差异均没有统计学意义(P>0.05)。结论染色时间和试剂浓度对细胞色度学参数影响明显,而对几何形态学参数无明显影响。
Objective The aim of this study was to explore concentrations on morphometric measurement of cells. Methods effect of varying staining times and dye Human erythrocytes ,were stained for 10, 20, and 40 min, and with varying ratio of solution A/B ( 1 : 1, 1 : 2, 1: 4) of Wright-Giemsa staining The parameters of colorimetry and geometric morphology of the erythrocytes were rested using image analysis software, and the data were compared and analyzed. Results There were statistical significances in red, green, blue three primary colors of tion groups (P 〈 0.05). The short time the erythrocytes among varying staining times and dye concentra- (10 min) and low dye concentration (1: 4) group had the lowest values of red, green, blue three primary colors, and the long time (40 min) and high dye concentration ( 1 : 1 ) group had the highest values of three primary colors (P 〈 0.05 ). There were no statistically signif- icant differences in area, perimeter, Conclusions Variations of staining and diameter of the erythrocytes among different groups (P 〉 0.05 ). time and dye concentration have significant influence on cell colorimetry measurement, but have no obvious effect on geometric morphology parameters
