摘要
目的探讨G蛋白抑制多肽汇利欣康(HLXK)对心肌肥大时microRNAs表达的影响。方法 36只成年♂昆明小鼠,随机分为对照组、模型组、HLXK组,每组12只。去甲肾上腺素(NE,1.5 mg·kg-1)腹腔注射造模,每日两次;HLXK组在造模同时给予HLXK(90μg·kg-1,ip,Bid);对照组注射给予等体积的生理盐水。在造模后d 20,称量小鼠体重和左心室重量,计算左心室重量指数。将消化培养的大鼠乳鼠心肌细胞接种于96孔板,分为对照组、模型组(NE 1μmol·L-1)、HLXK组(NE 1μmol·L-1+HLXK 11.30 mg·L-1),用[3H]-亮氨酸参入法检测蛋白质合成。以qRT-PCR测定心肌组织中microRNAs的表达。结果与模型组相比,HLXK治疗组小鼠左心室重量指数明显降低,心肌细胞肥大明显减轻,[3H]-亮氨酸参入量明显减少。NE可使小鼠心脏中和培养的大鼠心肌细胞中miR-199、miR-1-2-5p和miR-499的表达量明显增加,HLXK可明显抑制以上效应。结论 HLXK可抑制NE导致的小鼠心肌肥大,其机制可能与调节miR-199、miR-1-2-5p和miR-499的表达有关。
Aim To investigate the effects of G protein inhibitory peptide Huilixinkang(HLXK) on the expres-sions of microRNA in myocardial hypertrophy. Meth-ods 36 mice were divided into 3 groups, 12 mice in each group. The mice in control, model, HLXK groups were respectively administrated with normal sa-line, norepinephrine (NE) 1.5 mg .kg-l, NE 1.5 mg.kg-1 and HLXK 90 μg .kg-l(ip, bid). After 20 days treatment, the body weight and left ventricle weight of the mice were measured, and the left ventri-cle weight index was counted. The myocardial cells of newborn rats were divided into 3 groups, model and HLXK groups were treated with NE 1 μmol .L-1and NE 1 μmol .L-1 + HLXK 11.3 mg .L-1 The appli-cation of the[ 3H ]-leucine incorporation procedure wasassessed for measuring protein production ot myocarardial cells.Quantitivereverse transcription polymerase chain reaction (qRT-PCR) was performed to analyze the expression of microRNA in cardiac muscle and cul-tured myocardial cells. Results HLXK significantly inhibited left ventricle index and myocardial cell hyper-trophy, reduced [3H ]-leucine incorporation, de-creased the expression of miR-199, miR-1-2-5p and miR-499 in heart of mice and myocardial cells of new-born rats. Conclusion HLXK can alleviate myocardi-al hypertrophy induced by NE in mice; its mechanism may be related to the regulation of miR-199, miR-1-2- 5p and miR-499.
