红景天苷对诱导脐血间充质干细胞向肝样细胞分化的影响 被引量:4

Effect of Salidroside on differentiation of human umbilical cord blood mesenchymal stem cells into hepatocyte-like cells

摘要 目的研究红景天苷对诱导人脐血间充质干细胞(hUCB-MSCs)向肝样细胞分化的影响。方法体外分离培养hUCB-MSCs,分为红景天苷10μmol/L干预组(A组)和对照组(B组),观察诱导后细胞形态的变化,采用免疫荧光染色检测角蛋白18(CK18)的表达,ELISA法检测白蛋白(Alb)浓度,全自动生化分析仪检测丙氨酸氨基转移酶(ALT)水平。结果与B组相比,A组细胞形态变为三角形、多角形或类圆形,CK18、ALT、Alb表达均升高(P<0.01)。结论在红景天苷单独诱导作用下,hUCB-MSCs在体外能转化为肝样细胞。 Objective To investigate the effect of Salidroside on differentiation of human umbilical cord blood mesenchymal stem cells (hUCB-MSCs) into hepatocyte-like cells. Methods hUCB-MSCs were cultured in vitro and divided into two groups of A( treated with salidroside 10 μmol/L) and B(normal controls). The cell morphology was observed under inverted phase-contrast microscope. The expression of eeratin 18(CK18) was detected by immunofluorescent staining. The levels of albumin (Alb) and alanine aminotransferase (ALT) were detected by ELISA and fully automatic biochemical analyser, respectively. Results Compared with group B, hUCB-MSCs became triangular, polygonal and round, and the levels of CK18, ALT and Alb were increased in group A (P〈0. 01). Conclusion Salidroside in vitro can induce the hUCB-MSCs differentiating into hepatocytes-like cells.
