摘要
以海藻酸钠为载体,采用包埋交联法制备固定化α-淀粉酶,通过单因素试验和正交试验确定最优的固定化α-淀粉酶的条件。利用聚乙二醇4 000作为制孔剂,进一步制备具有多孔结构的固定化酶。结果表明,最优工艺为海藻酸钠1.6%,α-淀粉酶0.4%,羧甲基纤维素钠0.2%,CaCl22%,在此条件下,固定化α-淀粉酶的固定效率可达92.43%。在体系中添加浓度为2.5%的聚乙二醇4 000能使酶活性提高37.61个百分点,得到的固定化酶的温度稳定性以及pH稳定性均优于游离酶。
Using sodium alginate as carrier,α-amylase was immobilized by the embedding-crosslinking method.The optimal conditions were determined with the single factor experiment and the orthogonal experiment.The immobilized enzyme with porous structure was prepared by using polyethylene glycol 4 000 as the porogenic agent.The results showed that the optimal conditions were 1.6% sodium alginate,0.4% α-amylase and 0.2% sodium carboxymethyl cellulose,CaCl2 2%.Under the optimal conditions,the immobilization efficiency was 92.43%.The enzyme activity could be improved by 37.61 percentage with 2.5% PEG 4 000 in the optimized system.The temperature and the pH stability of the immobilized enzyme were found to be better than that of the free enzyme.
