摘要
[目的]以SAMP6及SAMR1鼠为研究对象,观察荣筋片对骨形态发生蛋白(BMP-2)、过氧化酶活化增生受体-γ(PPAR-γ)和脂联素(ADP)血清含量的影响,探讨荣筋片防治骨质疏松症的作用机制。[方法]选取6月龄SAMP6鼠36只,雌雄各半,随机分为SAMP6空白对照组、荣筋片组、仙灵骨葆组。另取11只同月龄性SAMR1鼠,作为同源对,将荣筋片配制成溶液。空白对照组给予0.5 mL生理盐水灌胃,每日1次,连续12周后。摘眼球采血,ELISA法检测血清BMP-2、PPAR-γ和ADP的含量。[结果]空白对照组BMP-2血清含量明显低于SAMRl对照组、仙灵骨葆和荣筋片组(P<0.01,P<0.05),差异有显著性。SAMRl对照组相比BMP-2血清含量高于仙灵骨葆组和荣筋片组(P<0.01,P<0.05)。而荣筋片组高于仙灵骨葆组(P<0.05)。空白对照组PPAR-γ血清含量明显高于SAMRl对照组(P<0.01)。仙灵骨葆组、荣筋片组和空白对照组组间没有统计学差异(P>0.05)。ADP血清含量空白对照组明显高于SAMRl对照组(P<0.01)。仙灵骨葆组和荣筋片组明显低于空白对照组(P<0.01),相比SAMRl对照组明显升高(P<0.01)。[结论]荣筋片与仙灵骨葆能增加SAMP6鼠血清促成骨分化因子BMP-2含量,降低SAMP6鼠血清促成脂分化因子PPAR-γ、ADP含量。两种中药可能改善自发性骨质疏松动物骨髓基质干细胞成骨-成脂分化的异常。
[Objective] Use senescence-accelerated mouse prone 6 （SAMP6） mice and senescence accelerated mouse re-istantl（ SAMRl） mice as the research object, observe the effection of Rong Jinpian on BMP-2, PPAR-γ and ADP in serum. [Methods] Choose 36 six months SAMP6 mices, male and female half, according to randomized SAMP6 blank control group, Rong jinpian group, Xianling Gubao group. Another 11 months with resistance to rapid aging SAMRI mice, as homologous comparison control group, the Rong Jinpian mixture solution according to the dose conversion. Blank control group to give 0.5 mL saline lavage, 1 times a day, 12 consecutive weeks. ELISA method to detect serum BMP- 2, PPAR-γ and ADP levels was used. [Results] Serum BMP-2 of blank control group was significantly lower than the other three groups （P〈0.01, P〈O.OS）. The SAMRI group was higher than the Xianling Gubao and Rong Jinpian group （P〈0.01, P〈0.05）. Serum PPAR-γ level of blank control group was significantly higher than the other three groups （P〈0.01, P〈0.05）. Compared with that blank control group, Rong Jinpian and Xianling Gubao group on ADP significantly lower. [Conclusion] Compared with SAMPI group, SAMP6 mice had lower BMP-2 level, higher PPAR-γ and ADP level in serum because spontaneous osteoporosis. Rong Jinpian and Xianling Gubao could raise BMP-2Ievel, dcrease PPAR-γ and ADP level in serum of SAMP6 mice.
