摘要
目的建立高效液相色谱法同时测定菊花中绿原酸、黄芩苷和槲皮素的含量。方法采用Hypersil ODSC18柱(4.6mm×250mm,5μm),甲醇-体积分数0.4%乙酸溶液为流动相,梯度洗脱,流速1.0mL·min-1,检测波长为337nm,柱温25℃。结果绿原酸、黄芩苷和槲皮素均能达到基线分离,且有较宽的线性范围和良好的线性关系(r〉0.9995);回收率均在99.2%-99.4%内。结论该方法简便准确,稳定好,可用于同时测定菊花中绿原酸、黄芩苷和槲皮素的含量。
Objective To establish an HPLC method for the determination of chlorogenic acid, baicalin, and quercetin in the Chrysanthemi Flos. Methods A hypersil ODS C18 column ( 4.6mm×250mm,5μm) was used with methanol-0. 4% acetic acid solution as mobile phase. Gradient elution programme was ap- plied with the flow rate of 1.0 mL. min-1. The detection wave length was set at 337 nm and the column temperature was 25 ℃. Results Chlorogenic acid, baicalin and quercetin were separated perfectly with wide linear range and good linear relationship ( r 〉 0. 999 5 ) ; average recovery were 99.2%-- 99.4%. Conclusions This method is simple and accurate, and the results are stable. It can be used to determine the contents of chlorogenic acid,baicalin and quercetin in Chrysanthemi Flos.
