对乙酰氨基酚琥珀酸酯在家兔体内的药代动力学研究

摘要 目的:研究肌肉注射对乙酰氨基酚琥珀酸酯在健康家兔体内的药代动力学。方法:采用RP-HPLC法测定6只成年健康家兔肌肉注射对乙酰氨基酚琥珀酸酯后不同时间点血浆中对乙酰氨基酚的浓度,应用DAS 2.0药动学软件处理血药浓度数据,拟合对乙酰氨基酚琥珀酸酯在家兔体内吸收模型,绘制药时曲线,计算药代动力学参数。结果:对乙酰氨基酚琥珀酸酯在家兔体内代谢符合一级吸收二室模型(权重系数=1/C2),主要药动学参数:Tmax=0.25h、Cmax=95.44mg/L、t1/2(α)=0.73h、t1/2(β)=2.70h、AUC=82.44mg/L*h、Vd=45.65L/kg。结论:与肌肉注射对乙酰氨基酚相比,家兔肌肉注射乙酰氨基酚琥珀酸酯的Tmax、t1/2(β)均缩短,表明乙酰氨基酚琥珀酸酯在体内起效迅速且消除较快。 Objective:To study the pharmacokinetics of acetaminophen succinate in healthy rabbit after intramuscular injection.Methods:RP-HPLC was used to determine the acetaminophen concentration of 6 adult healthy rabbits at different time after injecting acetaminophen succinate. Then the pharmacokinetic parameters were disposed by DAS 2.0 pharmacokinetic software.Results:The concentration-time curve of acetaminophen succinate in rabbit was single compartment model (weight coefficient=1/C2). The main pharmacokinetic parameters was:Tmax=0.25h, Cmax=95.44mg/L, t1/2(α)=0.73h, t1/2(β)=2.70h, AUC=82.44 mg/L*h, Vd=45.65L/kg.Conclusions:Compared with muscular injection of acetaminophen, the Tmax and t1/2(β) of muscular injection of acetaminophen succinate shorten obviously, which indicates that acetaminophensuccinateonsetquicklyandremoverapidly in vivo.
