摘要
目的 研究三种浓度的异丙酚对离体大鼠左右心室收缩功能的影响。方法 雄性SD大鼠 10只 ,经Langendorff离体心脏恒压灌注 ,依次梯度输入含有异丙酚 1、3和 10 μg/ml的灌注液。通过心室内的乳胶水囊经换能器连于MacLabInstruments和Macintosh微机系统 ,观察给药前后以及不同浓度异丙酚对左右心室等容收缩时压力和速度指标的影响。结果 10 μg/ml的异丙酚提高了左右心室的发展压 ,降低了舒张末期压 ;同时加快了左右心室收缩和舒张的速率 ;增加了冠脉灌流量。结论 异丙酚有保护心肌舒缩功能的作用 ,不直接抑制左右心室的内在收缩性 。
Objective The aim was to investigate the effects of propofol on left and right ventricular function. Methods Ten male rats were used. Measurements were made on isolated Langendorff perfused heart preparation. Developed pressure was measured from a fluid filled latex balloon in the right and left ventricle which was connected to MacLab instruments and Macintosh computer system through transducer. After 30-min equilibration time the isolated hearts were perfused with propofol 1,3 and 10 μg/ml in a stepwise fashion. Results With the increasing propofol concentration,left and right ventricular systolic and diastolic function did not change as indicated by continuous monitoring of developed pressure,dP/dtmax, dP/dtmin,Vpm,Vmax, end-diastolic pressure and T value. However propofol 10μg/ml increased the developed pressure and decreased end-diastolic pressure of both ventricles (P<0.05), moreover 10μg/ml propofol increased coronary flow significantly. Conclusion Propofol does not directly inhibit myocardial contractility of both left and right ventricle of rat heart. 10 μg/ml propofol can dilatate coronary artery in rats
